LncRNA PVT1调节SOCS2表达对胆管癌细胞增殖的影响

Abstract

目的:本研究旨在探究长链非编码RNA淋巴瘤易位变体1(lncRNA PVT1)和细胞因子信号抑制因子2(SOCS2)在胆管癌(CCA)细胞增殖和迁移中的作用,从而为CCA的发病机制提供新的理论见解。方法:收集2021年10月至2023年11月期间在承德医学院附属医院经手术确诊并经病理诊断的20例胆管癌患者的癌组织及癌旁组织样本。培养正常胆管上皮细胞(HIBEC)和胆管癌细胞系(RBE、HCCC9810、HUCCT1)。采用实时定量逆转录PCR(qRT-PCR)检测lncRNA PVT1的表达。将小干扰RNA(siRNA)转染入HUCCT1细胞以下调lncRNA PVT1的表达。通过细胞计数试剂盒8(CCK-8)检测细胞增殖情况,采用蛋白质印迹法(WB)分析胆管癌组织和细胞中SOCS2的表达水平,以及lncRNA PVT1敲低前后的情况。结果:qRT-PCR显示,CCA组织中 lncRNA PVT1的表达显著高于相邻组织(P<0.05)。在细胞系中,HUCCT1细胞中lncRNA PVT1的表达最高(与HIBEC相比,P<0.0001),且siRNA-PVT1-3的敲低效果最为显著(P<0.0001)。lncRNA PVT1的敲低显著降低了HUCCT1细胞的增殖(P<0.01)和迁移(P<0.0001)。WB 结果表明,CCA组织中SOCS2的表达低于正常组织(P<0.05),而敲低lncRNA PVT1后,SOCS2的表达得以恢复(P<0.0001)。结论:lncRNA PVT1在CCA中表达上调,通过抑制SOCS2促进肿瘤进展。沉默lncRNA PVT1可恢复SOCS2的表达,从而抑制CCA细胞的增殖。
