基于网络药理学探究穿心莲抗肝癌的作用研究

Abstract

目的:采用网络药理学方法探讨穿心莲抗肝癌的作用机制。方法:结合TCMSP数据库,确定穿心莲的活性成分和靶点;利用GeneCards、TTD、DisGeNET等数据库,对肝癌潜在的靶点进行预测,从两者中筛选出共有的靶标。利用STRING数据库和Cytoscape等分析平台,建立蛋白质互作网络。利用DAVID数据库,对共有靶标进行GO、KEGG的富集。结果:穿心莲中有24种有效成分和66个靶点,肝癌中有2655个不同的靶点,二者有51个交集靶点。蛋白相互作用网络拓扑分析显示,穿心莲治疗肝细胞癌的机制与TP53、IL6、AKT1、TNF等靶点有关。KEGG富集通路有122个,主要包括p53信号通路、PI3K/Akt信号通路、IL-17信号通路,且与癌症信号通路、乙型肝炎信号通路、丙型肝炎信号通路、非酒精性脂肪性肝病信号通路等等疾病信号通路高度相关。结论:穿心莲是一种多成分,多靶点,多途径的抗癌药物,为进一步探究穿心莲治疗肝癌的理论研究提供参考。
